UPLC-Q-TOF-MS法分析连花清瘟胶囊的入血成分

摘要:
[目的] 采用UPLC-Q-TOF-MS方法,首次对中成药连花清瘟胶囊经SD大鼠灌胃给药后吸收入血的成分进行分析。[方法] 采用大鼠眼眶后静脉丛取血,甲醇沉淀蛋白,采用C18色谱柱,以甲醇-0.1%甲酸水为流动相进行梯度洗脱,负离子模式采集,分别对空白血浆、混合对照品溶液及连花清瘟胶囊给药后血浆同时进行ULC-Q-TOF-MS分析。[结果] 根据色谱峰保留时间及质谱碎裂规律,结合文献和对照品的相关信息,确认了连花清瘟胶囊22个吸收入血原型成分,主要为黄酮、蒽醌、苯丙素和环烯醚萜类。[结论] 该研究初步明确了连花清瘟胶囊的入血原型成分,为后期体内代谢研究提供了前期的科学研究数据。

Analysis of absorbed components in rat plasma after oral administration of Lianhua Qingwen capsules by UPLC-Q-TOF-MS

Abstract:
[Objective] A UPLC-Q-TOF-MS method was developed for the analysis of absorbed components in SD rat plasma after oral administration of Lianhua Qingwen capsules (LQC) for the first time.[Methods] The blood samples were taken from the posterior orbital venous plexus of rats and methanol was used for the protein precipitation.The separation of components was achieved on a C18 column by gradient elution with methanol -0.1% formic acid water solution as mobile phase.Comparative analysis of blank plasma,mixed reference standards and plasma with LQC was performed by UPLC-Q-TOF-MS in negative ion mode.[Results] The results suggested that 22 prototype compounds were identified,including flavonoid,anthraquinone,phenylpropanoid and iridoid.These compounds were identified and confirmed by comparing their retention times,mass spectrometry fragmentation characteristics with that of references as well as literatures.[Conclusion] The study of the absorbed components of LQC in plasma provides an experimental basis for revealing the effective components and its pharmacokinetics study.
