补骨脂苷和异补骨脂苷与牛血清白蛋白相互作用研究

摘要:
[目的]研究补骨脂苷(PO)和异补骨脂苷(IPO)与牛血清白蛋白(BSA)的相互作用。[方法]通过紫外吸收光谱法、荧光淬灭光谱法、同步荧光光谱法、三维荧光光谱法分析PO和IPO对牛血清白蛋白的淬灭机制、结合常数、结合位点数及构象影响。[结果]PO和IPO对牛血清白蛋白产生了静态淬灭作用,两种药物与BSA有较弱的结合,在BSA上的结合位点均为位点Ⅱ,并且对BSA上色氨酸氨基酸残(Trp)基微环境产生了微弱的影响。[结论]借助光谱法揭示了PO和IPO与BSA相互作用的机制。

Abstract:
[Objective] To study of on the interaction between BSA and psoralenoside (PO) as well as isopsoralenoside (IPO).[Methods] Using the methods of UV absorption spectroscopy, fluorescence quenching spectroscopy, synchronous fluorescence spectroscopy and three-dimensional fluorescence spectroscopy quenching mechanism, binding constants, binding sites, conformational changes of PO as well as IPO to the BSA were analyzed.[Results] PO and IPO bound weakly to BSA with a static quenching process, which cause light changes in the polarity environment around the chromophore of tryptophan. The binding sites of the two drugs on BSA were Site Ⅱ.[Conclusion] The mechanism of interaction between PO and IPO with BSA was revealed through the spectroscopy study.
