基于网络药理学探讨红花治疗非酒精性脂肪肝的可能性

摘要:
[目的] 利用网络药理学探讨红花治疗非酒精性脂肪肝(NAFLD)的可能性。[方法] 通过中药系统药理数据库和分析平台(TCMSP)数据库得到红花OB≥30%,DL≥0.18的有效活动成分,通过Swiss靶点预测数据库获取成分对应靶点。利用DisGeNet数据库及OMIM数据库获取NAFLD的靶点。采用STRING数据库构建交互基因蛋白之间的关系网络,利用DAVID数据库分析GO富集和KEGG信号通路。[结果] 筛选得到红花的361个靶点,其中涉及NAFLD的靶点38个,主要与VDR、PPARD、TNF、INSR等靶点有关。KEGG富集分析结果表示红花治疗NAFLD与胰岛素抵抗、催乳激素信号通路、PPAR信号通路等通路有关。[结论] 研究通过网络药理学系统探讨了红花活性成分及其所对应的靶点,以及治疗NAFLD潜在通路,为深入研究红花治疗NAFLD的基础和临床研究提供参考。

To explore the possibility of Carthamus tinctorius in the treatment of non-alcoholic fatty liver based on the network pharmacology

Abstract:
[Objective] To study the mechanism of Carthamus tinctorius in the treatment of non-alcoholic fatty liver through network pharmacology.[Methods] The effective active components of Carthamus tinctorius OB ≥ 30% and DL ≥ 0.18 were obtained through the pharmacology database of traditional Chinese medicine system and analysis platform (TCMSP) database. The corresponding target of components were obtained through Swiss target prediction database. Targets of non-alcoholic fatty liver were obtained by using DisGeNet database and OMIM database.The relationship network between interactive gene proteins was constructed by using STRING database,and the GO enrichment and KEGG signal pathways were analyzed by using DAVID database.[Results] The 361 targets of Carthamus tinctoriu were screened,including 38 targets involving non-alcoholic fatty liver,which were mainly related to VDR,PPARD,TNF,INSR and other targets. The results of KEGG enrichment analysis showed that the treatment of non-alcoholic fatty liver by Carthamus tinctoriu was related to insulin resistance,prolactin signaling pathway,PPAR signaling pathway and other pathways.[Conclusion] In this study,the active components of Carthamus tinctoriu and their corresponding targets were discussed through the network pharmacology system,as well as the potential pathways for the treatment of non-alcoholic fatty liver disease,providing reference for the in-depth study of the basic and clinical research on the treatment of non-alcoholic fatty liver disease.
