中药心复康联合骨髓间充质干细胞移植对大鼠急性心肌梗死后心功能的影响*

摘要:

[目的]探讨中药心复康联合骨髓间充质干细胞(BMSCs)移植对大鼠急性心肌梗死后心功能的影响。[方法]经密度梯度离心和贴壁筛选法培养并纯化BMSCs,采用冠状动脉结扎法建立大鼠急性心肌梗死模型,术后采用中药联合干细胞移植,24 h检测血清超氧化物歧化酶(SOD)、丙二醛(MDA)活性,2周后超声心动仪检测大鼠心脏收缩功能[射血分数(EF)、左室短轴缩短率(FS)、心输出量(CO)];处死动物,硝基四氮唑蓝(N-BT)染色评价梗死面积。[结果]联合治疗组较单纯细胞移植组能明显减小大鼠心肌梗死面积,降低血中MDA水平,提高SOD水平,EF、FS、CO也有不同程度改善。[结论]中药联合骨髓间充质干细胞移植较单纯细胞移植对结扎大鼠左冠状动脉前降支所致的急性心肌梗死具有更明显的保护作用。

Abstract:

[Objective] To discuss the effect of XinFuKang (XFK) combining with Bone Mesenchymal stem cells (BMSCs) transplantation in treating acute myocardial infarction.[Methods] BMSCs were cultured and purified by gradient centrifugation-adhesive method.The acute myocardial infarction in rats was established by coronary artery ligation.The therapy method of XFK combined with BMSCs transplantation was implemented 15 minutes after the operation.Then the activity of serum SOD and MDA was examined in 24 hours.The examination of FE,FS,FO and N-BT was carried out two weeks after the operation.[Results] XFK combining with BMSCs transplantation therapy could reduce the infarct area,decrease serum MDA,enhance serum SOD,ameliorate EF,FS and CO.[Conclusion] Compared with BMSCs transplantation only,XFK combining with B-MSCs transplantation has a better protective effect against acute myocardial infarction.
