防哮灵胶囊对哮喘大鼠血清IFN-γ和IL-4含量的影响

摘要:
[目的]观察防哮灵胶囊对哮喘大鼠血清γ-干扰素(IFN-γ)和白介素-4(IL-4)含量的影响,探讨防哮灵胶囊治疗哮喘气道炎症的作用机制。[方法]清洁级雄性Wistar大鼠84只,随机分为7组,每组12只,采用卵蛋白制作大鼠哮喘模型。用酶联免疫吸附试验(ELISA)法检测各组大鼠血清中IFN-γ和IL-4的水平。[结果]B组血清中IFN-γ的含量明显低于正常组(P<0.05),而IL-4的含量明显高于前者(P<0.05),存在着严重的IFN-γ/IL-4比例的失衡;各治疗组均可不同程度的上调IFN-γ并下调IL-4的水平;其中尤以C组的作用显著,防哮灵胶囊各剂量组纠正失衡的IFN-γ/IL-4比例程度来看,随着给药剂量增加或提前干预作用趋势可加强。[结论]防哮灵胶囊治疗哮喘气道炎症的作用机制与纠正失衡的IFN-γ/IL-4水平有关。

Abstract:
[Objective] To observe the effect of FangXiaoLing capsule on serum IFN-γand IL-4 content in asthma rats and explore the effect mechanism of FangXiaoLing capsule in treating air passage of asthma.[Methods] Eighty-four Wistar rats were randomly divided into seven groups with 12 rats per group.The model of asthma in rats was established by using egg protein method.The serum IFN-γand IL-4 content of rats in each group were determined by ELISA method.[Results] The serum IFN-γcontent of B group was obviously lower than that of normal group (P<0.05).But serum IL-4 content of B group was obviously higher than that of normal group (P<0.05).An obvious unbalance between IFN-γ/IL-4 proportion was existed.All treatment group had up-regulation of IFN-γ and down-regulation of IL-4 level at different extent and the C group had the most obviously curative effect.It was noticed that the effect tendency of FangXiaoLing capsule may be strengthened obviously with the increasing of administration dosage or intervention in advance according to the change of the correction unbalance of IFN-γ/IL-4 extent.[Conclusion] The effect mechanism of FangXiaoLing capsule may be correlated with the correcting unbalance of IFN-γ/IL-4 ratio.
