虫草素抑制人胃癌细胞MKN-28运动侵袭能力研究

摘要:
[目的] 探讨虫草素对人胃癌细胞MKN-28体外迁移、侵袭能力的影响及其可能作用的分子机制。[方法] 悦悦运8法检测虫草素对MKN-28 细胞的增殖抑制作用,体外迁移、侵袭实验观察虫草素对人胃癌细胞MKN-28 迁移、侵袭能力的影响,反转录聚合酶链反应(RT-PCR)法检测虫草素作用24 h后MKN-28 细胞Ezrin 蛋白,基质金属蛋白9(MMP9)信使核糖核酸(mRNA)表达变化。[结果] 200~800 μg/mL 虫草素可明显抑制MKN-28 胃癌细胞增殖,体外迁移实验显示虫草素组细胞划痕闭合约18% ,对照组细胞划痕闭合约25% ,而表皮生长因子(EGF)组闭合约48% 。虫草素组显著低于另两组(P<0.05)。体外侵袭实验结果表明虫草素组穿过人工基底膜的细胞数明显减少(P<0.05);虫草素组Ezrin 和MMP9 的表达明显下调(P<0.05)。[结论] 虫草素可能通过下调Ezrin 及MMP9 表达抑制人胃癌细胞MKN-28 细胞的迁移与侵袭能力,从而发挥其抑制肿瘤生长的作用。

Effects of Cordyceps sinensis on the migration and invasion capability of human gastric adenocarcinoma cell line MKN-28

Abstract:
[Objective] To explore the effects of Cordyceps sinensis on the migration and invasion capability and on the expression of Ezrin and MMP9 in human gastric adenocarcinoma cell line MKN-28. [Method] The inhibitory effects of Cordyceps sinensis on the proliferation of MKN-28 cell line were evaluated by CCK-8 assay. The effects of Cordyceps sinensis on the migration and invasion ability of the cells were observed through scratch assay and Boyden chamber, respectively. RT-PCR system detecting the mRNA gene expression changes of Ezrin and MMP9 in MKN-28 cell line. [Result] Cordyceps sinensis from 200 μg/mL to 800 μg/mL can obviously inhibited the proliferation of the human gastric adenocarcinoma cell line MKN-28. Scratch and matrigel invasion experiments showed that cordyceps sinensis significantly inhibited cell migration and invasion. Cordyceps sinensis inhibited the gene expression of Ezrin and MMP9 in MKN-28 cell line. [Conclusion] Cordyceps sinensis may inhibited cell proliferation, migration and invasion through down regulating the expression of Ezrin and MMP9, then suppressing the tumor growth.
