[目的] 将大吞噬实验中的吞噬颗粒鸡红细胞换成酵母菌,硫代乙醇酸钠作为巨噬细胞刺激剂,观察小鼠腹腔巨噬细胞体内吞噬作用的现象和功能,探索和改进实验方法。[方法] 小鼠经腹腔注射3%硫代乙醇酸钠溶液刺激活化腹腔巨噬细胞,96 h后经腹腔注射分别注射鸡红细胞悬液和酵母菌菌液,10、30、60 min后观察巨噬细胞体内吞噬鸡红细胞和酵母菌的现象,并分别测定其吞噬百分率和吞噬指数。[结果] 30 min就可观察到小鼠腹腔巨噬细胞体内吞噬鸡红细胞和酵母菌的现象,吞噬鸡红细胞和酵母菌的平均吞噬百分率分别为15.3%和18.9%,吞噬指数分别为19.3和45.9,结果显示酵母菌作为吞噬颗粒,吞噬细胞对其吞噬百分数和吞噬指数均升高。[结论] 小鼠腹腔巨噬细胞体内吞噬酵母菌现象更明显,吞噬能力较大,而且省去制备鸡红细胞的时间,操作更方便,可作为一种稳定而简单体内巨噬细胞吞噬实验的方法。 |

The improved experiment of phagocytic function of peritoneal macrophages in mice |

[Objective] The use of saccharomycetes instead of chicken red blood cell in phagocytosis test, glucosinolates glycolic acid sodium as macrophages stimulants, observe the phenomenon and the function of phagocytosis of mouse peritoneal macrophages in vivo, and explore and improve the experiment method.[Methods] Mice were injected of 3% sodium thioglycolate solution by intraperitoneal induced macrophages in the peritoneal, chicken red blood cells suspension and saccharomycetes suspension were injected by intraperitoneal injection after 96 h, and 10, 30 and 60min later, observed the phenomenon of in macrophage phagocytosis of chicken red blood cells and saccharomycetes, and determine the phagocytic percentage and phagocytic index. [Results] The phenomenon of mouse macrophage phagocytosis of chicken red blood cells and saccharomycetes can be observed after 30 min, the mean phagocytic percentage of chicken red blood cells and saccharomycetes were 15.3% and 18.9% respectively, the phagocytic index of the chicken red blood cells and saccharomycetes were 19.3 and 45.9 respectively. The use of saccharomycetes as consuming particles can increase the phagocytic percentage and the phagocytic index.[Conclusion] The phenomenon of mouse peritoneal macrophage phagocytosis of saccharomycetes was more obvious in vivo, and the phagocytosis capacity is larger, but also save the time for preparation of chicken red blood cells, the operation was more convenient. |
