HPLC梯度洗脱法同时测定玉女煎多指标成分含量

摘要:
[目的]建立同时测定玉女煎中芒果苷、β-蜕皮甾酮、毛蕊花糖苷含量的高效液相色谱法。[方法]采用Ultimate XB-C18色谱柱(4.6 mm×250 mm,5 μm);以乙腈-0.1%磷酸溶液为流动相,梯度洗脱,流速:1.0 mL/min;检测波长:250 nm(芒果苷、β-蜕皮甾酮)和330 nm(毛蕊花糖苷)。[结果]芒果苷、β-蜕皮甾酮、毛蕊花糖苷质量浓度在10.0~400.0 μg/mL(r=0.999 9)、2.4~96 μg/mL(r=0.999 9)、2.0~80 μg/mL(r=0.999 9)范围内线性关系良好,3种成分的平均加样回收率(n=9)分别为100.2%、101.0%、98.1 8%,RSD分别为2.73%、1.95%、1.39%。[结论]该方法简便,重复性好,可为玉女煎的质量控制提供参考。

Abstract:
[Objection] To establish an HPLC method for simultaneous determina-tion of mangiferin,β-ecdysterone,verbascoside in Yunvjian.[Methods] The HPLC separetion was performed on Ultimate XB-C18 column (4.6 mm×250 mm,5 μm) eluted with the mobile phase as acetonitrile-0.1% phosporic acid solution in a gradient mode at a flow rate of 1.0 mL/min.The detection wavelength was 250 nm for mangiferin and β-ecdysterone,and 330 nm for verbascoside.[Results] The calibration curve was liner within the range of 10.0-400.0 μg/mL (r=0.999 9) for mangiferin,2.4~96.0 μg/mL (r=0.999 9) for β-ecdysterone,and 2.0~80.0 μg/mL (r=0.999 9) for verbascoside,and the average recovery were 100.2%,101.0%,98.1 8% with RSD were 2.73%,1.95% and 1.39%(n=9).[Conclusion] The method is simple and accurate,and can be used for the quality control of Yunvjian.
