杞黄颗粒对实验性脉络膜新生血管VEGF蛋白及其mRNA表达的影响

摘要:
[目的]探讨杞黄颗粒对激光诱导脉络膜新生血管(CNV)大鼠血管内皮生长因子(VEGF)蛋白及其mRNA表达的影响。[方法] 81只雄性棕色挪威(BN)大鼠,采用随机分组法,13只为空白对照组;68只造模,随机抽取3只验证CNV形成,剩余65只随机分为5组:杞黄颗粒A组(简称杞黄A组),杞黄颗粒B组(简称杞黄B组),雷珠单抗组,杞黄颗粒+雷珠单抗组(简称杞黄+雷珠组),模型组。除杞黄B组给药90 d,其余组给药60 d后检测VEGF蛋白含量和mRNA表达水平。[结果]造模第7天,CNV验证形成。用药60 d,杞黄+雷珠组与杞黄A组、模型组相比VEGF的蛋白含量也明显降低,均P<0.05,具有统计学差异。杞黄B组与杞黄A组相比,VEGF蛋白含量降低降低,P<0.05,具有统计学意义。用药60 d,空白对照组相比模型组,杞黄+雷珠组相比模型组VEGF mRNA均有明显降低,P<0.05,具有统计学差异;杞黄B组相比杞黄A组的VEGF mRNA降低明显,P<0.05,具有统计学意义。[结论]杞黄颗粒与雷珠单抗联合使用,对于VEGF的表达有明显的调节作用,能够减少CNV的形成,对湿性老年性黄斑变性(WAMD)有一定治疗意义。

Effect of Qihuang granule on experimental choroidal neovascularization VEGF Protein and its mRNA expression

Abstract:
[Objective] To investigate the effect of Qihuang granule on expression of vascular endothelial growth factor protein and its mRNA in laser-induced choroidal neovascularization.[Methods] Eighty-one male brown Norway rats were randomly divided into two groups:13 for the blank control group; 68 were irradiated with 532 laser light. Three rats among the irradiated rats were randomly selected to the form of cnv and others were randomly divided into 5 groups:Qihuang granule group A (Gavage 60 days), Qihuang granule group B (Gavage 90 days), lucentis group, Qihuang granule combine lucentis group and the model group,with different drugs and time of treatment respectively. Expect Qihuang granule group B rats gavaged for 90 days, Others with 60 days, all retinal pigment epithelium-choroid-sclera complex form were geted to detect the protein content and mRNA expression of VEGF.[Results] After modeled 7 days, suggestting CNV formated. after 60 days intervention, VEGF protein content change, Qihuang granule combine lucentis group compared with Qihuang granule group A,Qihuang granule group A combine lucentis group compared with the the model group,VEGF protein content was reduced, with statistical differences(P<0.05). Qihuang granule group B compared with Qihuang granule group A, VEGF protein content were decreased, with statistically significant after 60 days intervention(P<0.05), VEGF mRNA change:compared the blank group with the model group, Qihuang granule combine lucentis group compared with the model group, VEGF mRNA were decreased with statistical differences(P<0.05). Qihuang granule group B compared with Qihuang granule group A,VEGF mRNA were decreased with statistically significant(P<0.05).[Conclusion] The method of Qihuang granule combine lucentis, can regulate the expression of VEGF, reduce the formation of CNV, and has a certain therapeutic significance to wet age-related macular degeneration.
