白术中3种内酯成分的含量测定及指纹图谱研究 |

摘要: |
[目的] 建立白术中白术内酯Ⅰ、Ⅱ、Ⅲ的含量测定方法和指纹图谱方法,结合系统聚类分析研究不同产地白术饮片的质量。[方法] 建立高效液相色谱-紫外检测器(HPLC-UV)双波长测定白术内酯Ⅰ、Ⅱ、Ⅲ含量的方法和单波长测定指纹图谱的方法,对4个产地12批白术饮片进行了测定研究。[结果] 白术中白术内酯Ⅰ、Ⅱ、Ⅲ的含量测定方法线性关系良好,加样回收率合格。不同产地白术中白术内酯成分含量相差在1.9~3.8倍之间,3种内酯成分总量差异最大为2.5倍,其中3种内酯成分总含量最高的为浙江白术。不同产地白术饮片指纹图谱相似度较高,相似度在0.972~0.998之间。系统聚类分析结果与含量测定和指纹图谱分析结果具有较好一致性。[结论] 白术饮片中白术内酯的含量测定方法特征性强、操作简便,结合白术饮片指纹图谱与系统聚类分析可更好地控制白术的质量。 |

[Objective] To control the quality of Atractylodes macrocephala Koidz. from different provinces by determining the contents of AtractylenolideⅠ, Ⅱ, Ⅲ, establishing the fingerprint and clustering analysis.[Methods] The contents of AtractylenolideⅠ, Ⅱ, Ⅲ were determined on a HPLC-UV system at two wavelengthes and the fingerprint was established at a single wavelength. Twelve batches of Atractylodes macrocephala Koidz. from four different provinces were analyzed.[Results] The quantitative evaluation of AtractylenolideⅠ, Ⅱ, Ⅲ had good linear range, and the recovery were qualified. The content of AtractylenolideⅠ, Ⅱ, Ⅲ from different producing regions varied between 1.9~3.8 times. The content of atractylenolides in Atractylodes macrocephala Koidz. from Zhejiang was 2.5 times as the content of material from Anhui. The fingerprint of Atractylodes macrocephala Koidz. from different producting regions was highly similar. The similarity varied between 0.972~0.998. The results of hierarchical cluster analysis have good consistency with content determination and fingerprint analysis.[Conclusions] The characteristic method for the determining Atractylenolide Ⅰ, Ⅱ, Ⅲ and the fingerprint method based on holistic view combining with hierarchical cluster analysis were better to control the quality of the Atractylodes macrocephala Koidz. |
