白子菜总黄酮纯化物对急性高尿酸血症大鼠作用研究

摘要:
[目的] 考察白子菜总黄酮纯化物对大鼠急性高尿酸血症(HUA)的治疗作用。[方法] 将60只雄性Wistar大鼠随机分为6组:空白组、模型组、别嘌醇组、白子菜总黄酮低剂量组、白子菜总黄酮中剂量组、白子菜总黄酮高剂量组。采用氧嗪酸钾-次黄嘌呤联用法建立大鼠急性HUA模型。造模1 h后,别嘌醇组灌胃给予别嘌醇混悬液(25 mg/kg),白子菜总黄酮各给药组:低剂量组(45 mg/kg)、中剂量组(90 mg/kg)、高剂量组(180 mg/kg)分别灌胃给予相应浓度的白子菜总黄酮纯化物混悬液,每日1次,共7 d。每日记录大鼠体质量与食量变化。比较各组大鼠给药后血尿酸(BUA)、血肌酐(SCr)、血尿素氮(BUN)含量与腺苷脱氨酶(ADA)、肝脏黄嘌呤氧化酶(XOD)活性。将大鼠左肾经石蜡包埋后,苏木素-伊红(HE)染色,观察大鼠肾脏组织病理形态变化。[结果] 与空白组相比,模型组BUA显著升高(P<0.05)。与模型组相比,白子菜总黄酮低、中、高剂量组大鼠的BUA、BUN、SCr和XOD活性水平均显著降低(P<0.05),白子菜总黄酮低剂量组、中剂量组大鼠的ADA活性显著降低(P<0.05)。与模型组相比,白子菜总黄酮各给药组均可改善急性HUA模型大鼠的肾小球萎缩、肾小管扩张和肾间质纤维化情况。[结论] 白子菜总黄酮可显著降低大鼠HUA水平,减缓HUA造模剂造成的肝肾损伤,在HUA的治疗中有潜在的开发价值。

Study on the effect of total flavonoids purified from Gynura Divaricate on rats with acute hyperuricemia

Abstract:
[Objective] In order to investigate the therapeutic effect of total flavonoids purified from Gynura Divaricate on acute hyperuricemia(HUA) in rats.[Methods] Sixty male Wistar rats were randomly divided into 6 groups:blank group,model group,allopurinol group,low-dose group of total flavonoids of Gynura Divaricate,medium dose group of total flavonoids of Gynura Divaricate,and high-dose group of total flavonoids of Gynura Divaricate. The acute HUA model of rats was established by the combined use of potassium oxazinate and hypoxanthine. One hour after modeling,allopurinol group was given allopurinol suspension(25 mg/kg) by gavage, and all administration groups of total flavonoids of Gynura Divaricate:low dose group(45 mg/kg),medium dose group(90 mg/kg),high dose group(180 mg/kg) were given corresponding concentrations of total flavonoids of Gynura Divaricate extract suspension by gavage once a day for 7 days. The changes of body mass and food intake of rats were recorded daily. The contents of Blood Uric Acid(BUA),Serum Creatinine(SCR),Blood Urea Nitrogen(BUN) and the activities of Adenosine Deaminase(ADA) and liver Xanthine Oxidase(XOD) were compared. The left kidney of rats was embedded in paraffin and stained with Hematoxylin Eosin(HE) to observe the pathological changes of rat kidney.[Results] Compared with the blank group,BUA in the model group increased significantly(P<0.05). Compared with the model group,the activity levels of BUA,BUN,SCr and XOD of rats in the low,medium and high dose groups of total flavonoids of Gynura Divaricate were significantly lower(P<0.05),and the activity of ADA of rats in the low and medium dose groups of total flavonoids of Gynura Divaricate was significantly lower(P<0.05). Compared with the model group,each administration group of total flavonoids of Gynura Divaricate can improve the glomerular atrophy,renal tubular dilatation and renal interstitial fibrosis of acute HUA model rats.[Conclusion] The total flavonoids of Gynura Divaricate can effectively reduce the level of HUA in rats and slow down the liver and kidney injury caused by HUA modeling agent. It has potential development value in the treatment of HUA.
