随着分析技术的进步,基于高效液相色谱发展出二维液相色谱技术。二维液相色谱是将分离机制不同而又相互独立的两根色谱柱串联起来构成的分离系统。相比于传统的一维液相色谱,二维液相色谱具有峰容量大、特异性强、灵敏度高、分离能力强等优点,广泛应用于药品、食品等研究领域。中药成分复杂,许多成分结构相似、极性差异小,难以分离,传统的分析技术对于中药基质的研究具有一定局限性,二维液相色谱的出现使中药分析更加简便、快速、全面,推动了中药研究的进展。文章总结了二维液相色谱技术的概念、特点,以及对近年来不同分离模式的二维液相色谱在中药研究中的应用,为该技术今后的进一步发展与应用提供一定参考。 |

With the advancement of analytical techniques,two-dimensional liquid chromatography (2D-LC) was appeared based on high performance liquid chromatography. 2D-LC is a separation system which composed of two columns. The columns have different separation mechanisms and independent with each other. Compared with one-dimensional liquid chromatography,2D-LC has the advantages of large peak capacity,strong specificity,high sensitivity,and strong separation ability. 2D-LC has been widely used in pharmaceuticals,food,and other research fields. Due to the ingredients of traditional Chinese medicine(TCM) are very complex,many of them have similar structures and minor differences in polarity,which is difficult to be separated,traditional analytical techniques have some limitations. The invention of 2D-LC has made the analysis of TCM become more convenient,rapid,and comprehensive. It also promoted the development of TCM research. In this paper,the concept and feature of 2D-LC are summarized,and elaborated the application of 2D-LC in analysis of TCM in the past few years,and our work could provide a reference for the future development and application of 2D-LC. |
