[目的] 探讨青蒿素联合厄洛替尼对肺癌细胞的活性和凋亡的影响。[方法] 通过CCK-8实验检测青蒿素和/或厄洛替尼不同浓度处理下A549肺癌细胞的活性;流式细胞术检测青蒿素和厄洛替尼单独或联合作用下A549肺癌细胞凋亡行为变化;小鼠肿瘤内注射实验检测青蒿素和厄洛替尼单独或联合作用下对小鼠成瘤的影响。[结果] 青蒿素联合厄洛替尼作用A549肺癌细胞时细胞活性下降91%,最佳的用药浓度为10 μg/mL青蒿素和10 μg/mL厄洛替尼;青蒿素联合厄洛替尼诱导细胞凋亡,凋亡率高达90%,显著高于单独处理组;肿瘤内注射青蒿素和厄洛替尼,使小鼠体内肿瘤体积显著缩小。[结论] 青蒿素联合厄洛替尼能降低肺癌细胞活性,诱导其凋亡,也能有效地减少体内肿瘤体积。 |

Effects of artemisinin combined with erlotinib on lung cancer cells |

[Objective] To investigate the effect of artemisinin combined with erlotinib on the activity and apoptosis of lung cancer cells.[Methods] The activity of A549 lung cancer cells treated with artemisinin and erlotinib at different concentrations was determined by CCK-8. The apoptosis of A549 lung cancer cells was detected by flow cytometry. The effects of artemisinin and erlotinib were investigated by intratumoral injection mouse tumor.[Results] The cell viability of A549 lung cancer cells was decreased by 91% when combining artemisinin with erlotinib, and the optimal concentration was artemisinin at 10 μg/mL and erlotinib at 10 μg/mL. The apoptotic rate of the cells treated with artemisinin and erlotinib at optimal concentrations was up to 90%, which was signifcantly higher than that of the cells treated with artemisinin or erlotinib alone at the same concentration. The tumor volume of intratumoral injection of artemisinin and erlotinib was significantly smaller in mice.[Conclusion] Artemisinin combined with erlotinib can reduce lung cancer cell activity, induced apoptosis. In vivo can also effectively reduce the tumor volume. |
